Daniel’s Research Notes

How to measure hair thickness:

1. Microscope size calibration

To find the thickness, I started off by finding the scale, between pixels and lengths(micrometer). As shown above, I took a picture of a ruler, and inserted it into Fiji, also known as imageJ. Then, I counted the number of pixels in 3 millimeters, or 3000 micrometers, then we set the scale bar at 500 micrometers, in white on the picture. In other words, now we know the exchange ratio between pixels and micrometers.

2. Measurement

The pictures above are the strands of hair of subject #1 and subject #2, with the scale bar, which we found in the section above. The top hair is darker and thicker, while the bottom, which is dyed hair, is thinner and has a lighter tone. After finding and changing the pixels to micro meters, we can use the line, to find the width, or diameter of the strands, which came out as around 100 micro meters, and the second picture is around half the size. Therefore, after finding the exchange ratio, from pixels to either microns or um, we can find the width or diameter of a strand of hair.
